Oral small-molecule GLP-1 receptor agonist

Orforglipron

Also known as Foundayo, LY3502970

Approved medicine

Summary

Orforglipron is a once-daily oral GLP-1 receptor agonist that is a small molecule rather than a peptide, so it needs no injection and no fasting before dosing. The FDA approved it for chronic weight management in April 2026; in the ATTAIN phase 3 programme, participants on the highest dose lost 12.4% of body weight over 72 weeks.

How it works

Orforglipron binds the GLP-1 receptor at a different site from peptide agonists and is absorbed through the gut without the absorption enhancers that oral semaglutide requires.

Research use cases

Areas where Orforglipron has been studied. Inclusion here does not mean it works for that purpose.

  • Chronic weight management
  • Type 2 diabetes (phase 3)
  • Weight maintenance after injectable therapy (studied)

What the research found

Phase 2 obesity trial

Mean weight loss reached 14.7% at 36 weeks at the highest dose, versus 2.3% with placebo.1

Phase 3 type 2 diabetes (ACHIEVE-1)

In adults with early type 2 diabetes, orforglipron lowered HbA1c by 1.3 to 1.6 percentage points over 40 weeks, with dose-dependent weight loss.2

Phase 3 obesity (ATTAIN)

Over 72 weeks, mean weight loss was 12.4% on the highest dose among adherent participants, versus 0.9% on placebo. This supported the 2026 approval.3

Safety and open questions

Gastrointestinal effects similar to injectable GLP-1 agonists. Because it is not a peptide, it is not a reconstitution product, and no calculator is provided.

Regulatory status

FDA-approved for chronic weight management (Foundayo, April 2026). Prescription only.

Status was accurate when this page was last reviewed on October 26, 2026. Regulations change; check with the relevant authority before relying on it.

Orforglipron: common questions

Is orforglipron a peptide?

No. It is a non-peptide small molecule that activates the GLP-1 receptor. We include it because it is part of the same research story and is often compared with peptide GLP-1 drugs.

Is Orforglipron approved for human use?

FDA-approved for chronic weight management (Foundayo, April 2026). Prescription only.
